What Jakavi is and what it is used for<\/h2>\n\n\n\n

Jakavi contains the active substance ruxolitinib.<\/p>\n\n\n\n

Jakavi is used to treat adult patients with an enlarged spleen or with symptoms related to myelofibrosis, a rare form of blood cancer.<\/p>\n\n\n\n

Jakavi is also used to treat patients with polycythemia vera who are resistant to or intolerant to hydroxyurea.<\/p>\n\n\n\n

How Jakavi works<\/strong><\/p>\n\n\n\n

Enlargement of the spleen is one of the hallmarks of myelofibrosis.\u00a0Myelofibrosis is a disorder of the\u00a0bone marrow, which means that the marrow is replaced by scar tissue.\u00a0The abnormal\u00a0bone marrow\u00a0can no longer produce enough normal\u00a0blood cells, which leads to a significant increase in the size of the spleen.\u00a0By blocking the activity of certain\u00a0enzymes\u00a0(called Janus kinases), Jakavi can reduce the size of the spleen in patients with myelofibrosis and relieve symptoms such as fever, night sweats, skeletal pain, and weight loss in patients with myelofibrosis.\u00a0Jakavi can help reduce the risk of serious blood or vascular complications.<\/p>\n\n\n\n

Do not take Jakavi<\/strong><\/p>\n\n\n\n

  • if you are allergic to ruxolitinib or any of the other ingredients of this medicine (listed in section 6).<\/li>
  • if you are pregnant or breastfeeding.<\/li><\/ul>\n\n\n\n

    If any of the above applies to you, tell your doctor who will then decide if you should start treatment with Jakavi.<\/p>\n\n\n\n

    Warnings and cautions<\/h2>\n\n\n\n

    Talk to your doctor or pharmacist before taking Jakavi<\/p>\n\n\n\n

    • if you have an infection . It may be necessary to treat your infection before you start taking Jakavi. It is important that you tell your doctor if you have ever had tuberculosis or if you have been in close contact with someone who has or has had tuberculosis . Your doctor may take samples to see if you have tuberculosis or other infections . It is important that you tell your doctor if you have ever had hepatitis B.<\/li>
    • if you have kidney problems. Your doctor may need to give you another dose of Jakavi.<\/li>

      • if you get a fever, chills or other symptoms of infection you.<\/li>
      • if you get a chronic cough with blood-stained cough, fever, night sweats and weight loss (these may be signs of tuberculosis ).<\/li>
      • if you have any of the following symptoms or if someone close to you notices that you have any of these symptoms: confusion or difficulty thinking, loss of balance or difficulty walking, clumsiness, speech difficulties, decreased strength or weakness in one side of the body, blurred and \/ or or loss of vision. This may be a sign of a serious brain infection and your doctor may suggest further examinations and follow-ups.<\/li>
      • if you develop painful skin rash with blisters (this is a sign of shingles ).<\/li>
      • if you notice skin changes. This may require further observation, as certain types of skin cancer (non- melanoma type) have been reported.<\/li><\/ul>\n\n\n\n

        Blood tests<\/p>\n\n\n\n

        Before starting treatment with Jakavi, your doctor will examine your blood to determine the best starting dose for you.\u00a0You will need to provide more blood samples during treatment so that your doctor can check the number of\u00a0blood cells\u00a0(\u00a0white blood cells,\u00a0red blood cells,\u00a0and\u00a0platelets\u00a0) in your body, and assess how you respond to the treatment and whether Jakavi has an unwanted effect on the\u00a0blood cells.\u00a0Your doctor may need to change your\u00a0dose\u00a0or stop your treatment.\u00a0Your doctor will carefully check if you have any signs or symptoms of\u00a0infection\u00a0before and during treatment with Jakavi.\u00a0The doctor will also regularly check the level of lipid\u00a0is (fat) in the blood.<\/p>\n\n\n\n

        To stop with Jakavi<\/p>\n\n\n\n

        When you stop taking Jakavi, the myelofibrosis symptoms may return. Your doctor may want the amount of Jakavi taken each day to be reduced gradually, before stopping completely.<\/p>\n\n\n\n

        Children and young people<\/h2>\n\n\n\n

        This medicine is not intended for use by children or adolescents under 18 years of age as it has not been studied in this age group.<\/p>\n\n\n\n

        Other medicines and Jakavi<\/h2>\n\n\n\n

        Tell your doctor or pharmacist if you are taking, have recently taken, or might take any other medicines.<\/p>\n\n\n\n

        You must mention the following types of medicines that contain any of the mentioned active substances as your doctor may need to change your\u00a0dose\u00a0of Jakavi.<\/p>\n\n\n\n

        The following may increase the risk of side effects of Jakavi:<\/p>\n\n\n\n

        • Some medicines used to treat your infection . These include medicines used to treat fungal infections (such as ketoconazole , itraconazole, posaconazole, fluconazole and voriconazole), medicines used to treat certain types of bacterial infections ( antibiotics such as clarithromycin, telithromycin, ciprofloxacin or erythromycin), including HIV infection \/ AIDS (such as amprenavir, atazanavir, indinavir, lopinavir \/ ritonavir, nelfinavir, ritonavir, saquvinavir), medicines for the treatment of hepatitis C (boceprevir, telaprevir).<\/li>
        • Nefazodone, a medicine used to treat depression.<\/li>
        • Mibefradil or diltiazem , medicines used to treat hypertension and chronic angina.<\/li>
        • Cimetidine, a medicine used to treat heartburn.<\/li><\/ul>\n\n\n\n

          The following may reduce the effect of Jakavi:<\/p>\n\n\n\n

          • Avasimib, a medicine used to treat heart disease.<\/li>
          • Phenytoin , carbamazepine or phenobarbital and other drugs for epilepsy that is used to stop seizures.<\/li>
          • Rifabutin or rifampicin, a medicine used to treat tuberculosis (TB).<\/li>
          • St. John’s wort ( Hypericum perforatum<\/em> ), a (traditional) herbal medicine used to treat mild depression.<\/li><\/ul>\n\n\n\n

            While taking Jakavi<\/strong>, never start taking a new medicine without first consulting the doctor who prescribed Jakavi.\u00a0This includes prescription drugs, over-the-counter drugs, herbal remedies, and alternative medicines.<\/p>\n\n\n\n

            Pregnancy and breastfeeding<\/h2>\n\n\n\n

            Do not take Jakavi during pregnancy. Talk to your doctor about the steps you should take to avoid getting pregnant during treatment with Jakavi.<\/p>\n\n\n\n

            Do not breastfeed while taking Jakavi.\u00a0Tell your doctor if you are breastfeeding.<\/p>\n\n\n\n

            Driving and using machines<\/h2>\n\n\n\n

            If you feel dizzy after taking Jakavi, do not drive or use machines.<\/p>\n\n\n\n

            Jakavi contains lactose and sodium<\/h2>\n\n\n\n

            Jakavi contains lactose (milk sugar). If you have been told by your doctor that you have an intolerance to some sugars, contact your doctor before taking this medicine.<\/p>\n\n\n\n

              During treatment, your doctor may recommend a lower or higher dose for you if the results of the blood tests show that this is necessary, if you have problems with your liver or kidneys or if you also need treatment with other medicines.<\/p>\n\n\n\n

              If you are on\u00a0dialysis, you should either take a single\u00a0dose\u00a0or two separate\u00a0doses\u00a0of Jakavi on your dialysis days, after one\u00a0dialysis\u00a0has been performed.\u00a0Your doctor will tell you if you should take one or two\u00a0doses\u00a0and how many tablets to take for each\u00a0dose.<\/p>\n\n\n\n

              Like all medicines, this medicine can cause\u00a0side effects, although not everybody gets them.<\/p>\n\n\n\n

              Most of Jakavi’s side effects are mild to moderate and generally disappear after a few days to a few weeks of treatment.<\/p>\n\n\n\n

              Tell your doctor if you experience any of the following\u00a0side effects.\u00a0Some are very common (may affect more than 1 in 10 people), some are common (may affect up to 1 in 10 people):<\/p>\n\n\n\n

              • signs of bleeding in the brain, such as sudden changes in levels of consciousness, persistent headache, numbness, tingling, weakness or paralysis (common)<\/li>
              • signs of bleeding in the stomach or intestines, such as black or bloody stools or bloody vomiting (very common)<\/li>
              • unexpected bruising and \/ or bleeding, abnormal fatigue, shortness of breath during exercise or at rest, unusually pale skin or frequent infections (possible symptoms of blood disease) (very common)<\/li>
              • painful rash with blisters (possible symptoms of shingles ( herpes zoster<\/em> )) (very common)<\/li>
              • fever, chills or other symptoms of infection (very common)<\/li>
              • low levels of red blood cells ( anemia<\/em> ), low levels of white blood cells ( neutropenia<\/em> ) or low levels of platelets ( thrombocytopenia<\/em> ) (very common)<\/li><\/ul>\n\n\n\n

                      Keep this medicine out of the sight and reach of children.<\/p>\n\n\n\n
